My thesisi project investigates the effect of a disintegrin and metalloprotease (ADAM), ADAM17, on natural killer (NK) cell antibody mediated cellular cytotoxicity (ADCC) in an ovarian cancer model. One of ADAM17s substrates, which it cleaves from the cell surface, CD16 is important for presenting antibodies to targeted cells - a vital step in ADCC. We have altered and introduced a non-cleavable form of CD16 into stem cells which we have differentiated into NK cells. Preliminary in vitro studies suggest NK cells with non-cleavable CD16 have enhanced killing capabilities. We are pursuing in vitro studies using a mouse model of ovarian cancer.
